You must do this until you’ve hit 20,000 skill points without the addition of whatever multiplier is next to it. If you crash or stop driving, you will break your skill point chain and have to start over.

Sometimes a challenge will ask you to obtain an ultimate skill chain in a specific area, such as Dunas Blancas. In this case, you must complete the challenge in the specified location. In Dunas Blancas, this means flying over sand dunes and hoping not to flip before accumulating 20,000 skill points.
